Genetic transformation of Paulownia elongata S. Y. Hu., mediated by Agrobacterium tumefaciens and biolistic system

Objective: The most appropriate conditions for genetic transformation through direct (bioballistic) and indirect (Agrobacterium tumefaciens) systems in Paulownia elongata were established.
 Design/methodology/approach: Starting from vitro propagation both organogenesis, internodal stem segments with 0.5 to 1 cm length determined as the best explant. optimum dose selection media was be 15 mg L-1 of kanamycin. It possible obtain transgenic plants under systems. In case Agrobacterium tumefaciens, two hours incubation, 48 h co-cultivation, optical density 0.9 used; while bioballistics, 120 PSI shot pressure, height at level 6 (16 cm), vacuum pressure 22 Hg mm, particle inflow gun system (PIG).
 Results: Both produced complete transformants, chimeras, well those confirmed by histochemical X-GLUC PCR analysis, producing a total 14 positive A. tumefaciens 26 trials ten bioballistic 30 trials; construction chitinase glucanase, NPT II gene GUS reporter used.
 Findings/conclusions: So far, this has been first report including integration glucanase genes.
